Synthesis and Evaluation of Phenyl-Thiourea Derivatives with Potential Antibacterial Effects

Background: We decided to build new derivatives using benzocaine with more potency to prevent drug resistance, in which hydrazone and thiourea moieties were combined in one structure, have been synthesized starting from 4-amino-phenyl-ethyl ester (benzocaine) for evaluation of their anti-bacterial potency.

Methods: 4-amino phenyl ethyl ester 2 as a base compound was produced by known methods from benzoic acid 1. 4-amino phenyl hydrazide 3 was prepared by reaction of hydra- zine hydrate with compound 2. 4-amino phenyl hydrazone 4, 5 were prepared by reaction of compound 3 with 3- nitrobenzaldehyde and 3-pyridine carbaldehyde. Target com-pounds were synthesized by the reaction of compound 4, 5 with corresponding aliphatic isothiocyanat.

Results: Thin layer chromatography was used to access the reaction and purity of the compounds synthesized. The melt-ing points were determined in open capillary tubes and pre-sented uncorrected. …
