Kinetic Study of Bacteriocin Produced by Isolated Lab on Gram Positive and Gram Negative Pathogenic Bacteria

Background: The objectives of this study are screening bac- teriocinogenic LAB from traditional dairy products and ki- netic study of bacteriocins on the selected spoilage and path- ogenic microorganisms.

Methods: The agar diffusion bioassay was used to initiate screening of isolated lactic acid bacteria from traditional dairy products for bacteriocin against standard gram positive and gram negative bacteria (Escherichia coli (PTCC= 1330), Micro- coccus luteus(PTCC= 1110), Staphylococcus aureus(PTCC= 1112), Bacillus cereus(PTCC= 1015), Listeria monocyto- genes(PTCC=1306)). Out of seventy nine LAB isolates, thir- ty four were found to inhibit these bacteria and Micrococcus luteus was the most sensitive one, then Producers were select- ed to obtain the kinetic changes on growth curve of indicator organisms by turbidometric assay.

Results: The comparison of the bacteriocins inhibition effect on indicator growth curves showed the different effect based on the type of bacteriocin and the indicator. …
