Cloning and Expression of Tnf Alpha Receptor in E.coli

Article excerpt

Background: TNF alpha is an adipokine involved in system- ic inflammation and it is a member of a group of cytokines that stimulate the acute phase reaction. It is produced by macrophages and different cell types such as CD4+ lympho- cytes, NK cells.Deregulation of TNF production has been implicated in a variety of human diseases such as arthritis rheumatoid. Recombinant TNF alpha receptor helps to con- trol and treatment arthritis rheumatoid symptoms.

Methods: In this study, TNR sequence was synthesized based on NCBI genbank and cloned into PGH vector. The construct was transformed to E.coli strain BL21 and ex- pressed in optimal conditions.

Results: The TNF receptor fragment in pGH vector was confirmed by PCR and restriction analysis. …
