Stable Transferring of Mutant Streptokinase Gene into E. Coli

Background: In this research we used mutant Streptokinase lack 42 C-terminal amino acids that has shown to lower levels of antigenicity.

Methods: Two sequences from Arabinose operon was am- plified by PCR using specific primers with appropriate re- striction enzyme sites in their end. These genes were cloned in pBluescriptIISK(+/-). Then synthetic mut - C42 Strepto- kinase under PLP promoter was cloned. In future it will be transferred between two homologous arms in pBluescript. This construct will be transformed to E. coli (HB101) genome by homologous recombination, after that 420C heat will stimulate expression of Streptokinase.

Results: Cloning of two homologous arms were confirmed by colony PCR, cloning of PLP promoter was confirmed by enzymatic digestion. After transferring this construct into E. …
