DETECTION OF TOXOPLASMA GONDII OOCYSTS IN SOIL OF URBAN PARKS, BASED ON MOLECULAR AND STAINING TECHNIQUES (ARAK, Iran)

Background: In this study, soil contamination of public parks in the Arak city with Toxoplasma gondii oocysts was examined.

Methods: Sixty soil samples were collected from 15 main parks of Arak city. Samples were taken from 4 places at each public parks. The site includes a potting area, around toilets, children's play area and the place of rest. The samples were transported to the Laboratory of Parasitology, Medical Sci- ences, and were dried at laboratory temperature. Floatation in saturated sucrose was performed for isolation of oocysts from the soil samples. Floating debris was tested by two methods: staining by the modified Ziehl-Neelsen technique and polymerase chain reaction.

Results: From 60 soil samples of public parks of Arak city, 8 samples (13%) were suspected to be contaminated with Tox- oplasma oocyst by staining method. But, only 3 samples (5%) of 60 samples were positive in PCR. …
